Captain Andrew Strauss has 19 and Alastair Cook is on 20 at Headingley, Leeds.
Earlier, the visitors were all out for 419, having resumed on 262-5 today after being put in to bat. Alviro Petersen scored 182 before he was caught behind off the bowling of Stuart Broad.
JP Duminy was 48 not out after captain Graeme Smith scored 52 and AB de Villiers 47. Broad had 3-96, while Steven Finn and James Anderson each took two wickets.
Play ended early at about 5:15 p.m. because of bad light.
England trails 1-0 in the three-game contest. South Africa can replace the hosts at the top of the International Cricket Council Test rankings by winning the series.
